Artificial insemination (AI) has been accepted as the primary breeding tool in genetic upgrading programmes in Sri Lanka.AI technique has been used to raise the genetic potential of cattle by using genetically superior bulls. Use of sexed semen for AI is now a reality for many cattle breeders.Sperm sexing with density gradient is more appropriate to Jaffna Peninsula because of the lower cost and without damages to sperm viability. Thus the present study was undertaken to improve the quality of AI service in Northern Province and separate X and Y-bearing bovine sperm by using a discontinuous sucrose density gradient. In conclusion the discontinuous sucrose density gradients can be considered as low cost tool for sperm sexing of bovine semen.
