Cyclosporine A reverses immunoendocrine damages induced by ketoconazole

In this study, we based in the utilization of 2 treatments (Ketoconazole and the Cyclosporine A) and 3 behavioral tests (Plus maze, Morris Water Maze and Forced Swimming). Ketoconazole (KTCZ), an orally active antifungal, was chosen in our study, with its neuro-behavioral properties (antidepressant and anxiolyic) and physiological (inhibitory effect on ganads and adrenals). Cyclosporine (CsA), a powerful immunosuppressant had a significant impact on transplant medicine. Its molecular action has been well defined in T cells and involved inhibition of critical signalling pathways that regulated T-cell activation. In this study, we demonstrated that the treatment of male Wistar rats by high dose ketoconazole causes anxiolytic and antidepressant-like behavior. Nevertheless these behavioral effects are associated with testicular damage, adrenal and thymus. Co-administration of cyclosporin A (CsA) restore partially or completely the domages.
